Abstract
The invention relates to methods and substances for the detection of thiopurine methyltransferase polymorphisms (TPMT). The method for the detection of TPMT polymorphisms comprises the following sequential steps: (i) providing a biological sample containing patient DNA; (ii) providing a first mixture containing primers and two hydrolysis probes adapted for the amplification of the DNA target sequence and the detection of the TPMT gene polymorphism rs1800462 (TPMT*2); (iii) providing a second mixture containing primers and two hydrolysis probes adapted for the amplification of the DNA target sequence and the detection of the TPMT gene polymorphism rs1800460 (TPMT*3B); (iv) providing a third mixture containing primers and two hydrolysis probes adapted for the amplification of the DNA target sequence and the detection of the TPMT gene polymorphism rs1142345 (TPMT*3C); (v) providing a polymerase chain reaction (PCR) premix; (vi) adding DNA extracted from the biological sample and the premix to the first, second and third mixtures for performing quantitative PCR and amplifying the target nucleic acid; (vii) measuring fluorescence and determining TPMT gene polymorphisms by comparing the amplification results with previously known samples.
